Raman spectroscopy is continuously confirming its potentials as a powerful tool in medical diagnosis, also thanks to multivariate analysis for extracting quantitative information from the noisy and spectrally-reach Raman spectra usually obtained from biological tissues and humors1. Unfortunately, multivariate analysis methods require very large amount of data, thus resulting hardly appropriate for diagnosis or monitoring rare pathologies for which a small number of clinical cases (and independent Raman spectra) are available. Pemphigus Vulgaris (PV) is a rare life-2threatening autoimmune disease, which induces significant changes in micro-Raman 3spectra of biopsies from ill patients. In order to exploit the information embedded in Raman spectra for PV therapy monitoring, it’s crucial to overcome the limitation due to the availability of few cases. To this aim, a linear or univariate correlation approach is here built and employed for the analysis of Raman spectra from oral tissues of PV affected patients, at different illness stages.
